The hospital is located in Main Road in Tsolo, Eastern Cape. Main Road is also known as the R396 and runs through the town of Tsolo. The R396 is off the N2 Highway between Kokstad and Mthatha.

Dr Malizo Mphele Hospital is a provincial hospital, and services are free to pregnant and breastfeeding women as well as children under the age of six years old. Other citizens are charged according to a sliding scale depending on their income.
